4-year Survival Outcomes in the dMMR/MSI-H Subpopulation of RUBY Part 1

The 4-year follow-up analysis demonstrated sustained long-term PFS benefit with JEMPERLI + CP vs placebo + CP*1

Descriptive analysis (not statistically powered) of dual primary endpoint: 4-year follow-up of PFS in the dMMR/MSI-H population (N=118)

Only 4 new progression events with an additional 2.5 years of follow-up from IA1, with median PFS not reached in the JEMPERLI + CP arm

*Median expected follow-up, 55.6 months (range, 49.9–67.7).1

CI=confidence interval; CP=carboplatin + paclitaxel; dMMR=mismatch repair deficient; HR=hazard ratio; IA1=first interim analysis; mPFS=median progression-free survival; MSI-H=microsatellite instability-high; NE=not estimable; PFS=progression-free survival.

In the JEMPERLI + CP arm, patients who were progression-free at 2 years had >90% probability of remaining progression-free and alive at 4 years1

Conditional PFS in the dMMR/MSI-H population (n=53)1

dMMR/MSI-H population JEMPERLI + CP
3-year progression-free probability from 1-year landmark 88.6%

dMMR/MSI-H population JEMPERLI + CP
3-year survival probability from 1-year landmark 84.0%

Conditional survival is an alternative and dynamic estimate of survival, representing the probability that a patient remains free of progression and death (cPFS) or remains alive (cOS) for additional time from a specified time point.1

CI=confidence interval; cOS=conditional overall surivival; CP=carboplatin + paclitaxel; cPFS=conditional progression-free survival; dMMR=mismatch repair deficient; MSI-H=microsatellite instability-high; OS=overall survival; PFS=progression free survival.

RUBY Part 1 Trial Design: A phase 3, randomised, double-blind trial of patients with primary advanced or recurrent EC (N=494, all-comers) who were randomised 1:1 to JEMPERLI + CP or placebo + CP Q3W for 6 cycles, followed by JEMPERLI or placebo Q6W, respectively, until disease progression, unacceptable toxicity, or up to 3 years. Major efficacy endpoints were investigator-assessed PFS by RECIST v1.1 in the dMMR/MSI-H and all-comers populations, and OS in all-comers.2

dMMR=mismatch repair deficient; EC=endometrial cancer; MSI-H=microsatellite instability-high; Q3W=every 3 weeks; Q6W=every 6 weeks; RECIST v1.1=Response Evaluation Criteria in Solid Tumours v1.1.

JEMPERLI Is Indicated

  • in combination with carboplatin and paclitaxel for the first-line treatment of adult patients with primary advanced or recurrent endometrial cancer (EC) and who are candidates for systemic therapy.
  • as monotherapy for the treatment of adult patients with mismatch repair deficient (dMMR)/microsatellite instability-high (MSI-H) recurrent or advanced EC that has progressed on or following prior treatment with a platinum-containing regimen.
